品系描述:重(zhong)(zhong)(zhong)組(zu)(zu)激活基因(yin)(yin)1(Recombination-activating gene 1,Rag1)編碼Rag1重(zhong)(zhong)(zhong)組(zu)(zu)酶,其在(zai)催(cui)化V(D)J重(zhong)(zhong)(zhong)組(zu)(zu)過(guo)程(cheng)中(zhong)發揮重(zhong)(zhong)(zhong)要(yao)的(de)(de)作用。V(D)J重(zhong)(zhong)(zhong)組(zu)(zu)過(guo)程(cheng)中(zhong)發生(sheng)的(de)(de)免疫球蛋(dan)白(Ig)基因(yin)(yin)和T細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)受體(TCR)基因(yin)(yin)的(de)(de)重(zhong)(zhong)(zhong)排(pai)和重(zhong)(zhong)(zhong)組(zu)(zu)是B細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)T淋巴細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)成(cheng)(cheng)熟過(guo)程(cheng)中(zhong)的(de)(de)必(bi)需階段。 Rag1敲除(chu)純(chun)合小(xiao)鼠導(dao)致VDJ重(zhong)(zhong)(zhong)組(zu)(zu)不(bu)能正(zheng)常(chang)進行,無法產生(sheng)成(cheng)(cheng)熟的(de)(de)T細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)和B細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)。與Prkdc缺(que)失(shi)(shi)的(de)(de)Scid模型(xing)不(bu)同(tong),Rag1敲除(chu)純(chun)合小(xiao)鼠導(dao)致所(suo)有成(cheng)(cheng)熟的(de)(de)淋巴細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)缺(que)失(shi)(shi)更徹底,且不(bu)造成(cheng)(cheng)“泄露(lu)(Leak)”。Rag1敲除(chu)小(xiao)鼠無CD3+T細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)受體(T Cell Receptor,TCR)αβ+陽(yang)性細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao),純(chun)合突變小(xiao)鼠體內(nei)胸腺細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)數(shu)量遠低于相應雜合子、野生(sheng)型(xing),其胸腺細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)主要(yao)為(wei)CD8-CD4-和IL2RG+,脾臟和骨髓中(zhong)缺(que)失(shi)(shi)IgM/IgD細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao),缺(que)失(shi)(shi)成(cheng)(cheng)熟的(de)(de)B細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)。實驗數(shu)據顯示Rag1敲除(chu)小(xiao)鼠體內(nei)B細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)和T細(xi)(xi)(xi)(xi)(xi)(xi)(xi)(xi)胞(bao)(bao)(bao)(bao)(bao)的(de)(de)發育停滯在(zai)早期階段。
